Current management of osteosarcoma relies heavily on standard multimodal strategies, including surgery combined with traditional chemotherapy agents like methotrexate and doxorubicin. However, the approved therapeutic landscape remains restricted, with few options specifically tailored for the disease. The emergence of novel assets is beginning to reshape this environment, shifting the focus toward personalized mechanisms of action.
Key candidates driving this evolution include OS Therapies' OST-HER2, a HER2-targeted immunotherapy currently in Phase IIb trials, and Iterion Therapeutics' tegavivint, which targets the Wnt/β-catenin signaling pathway. Other promising agents include SERB Pharmaceutical's DANYELZA, MedPacto's vactosertib, and GSK's Risvutatug rezetecan, an antibody-drug conjugate targeting B7-H3. Despite this progress, drug development remains concentrated in early-stage clinical trials. Experts note that while research into tyrosine kinase inhibitors and cell-based therapies is accelerating, the lack of large-scale, late-stage data continues to pose a challenge for regulatory approval and consistent global availability. Future market success will largely depend on whether these emerging candidates can demonstrate statistically significant improvements in overall survival and durable response rates in broader patient populations.
